The dissertation is devoted to the investigation of the energy-dependent
Ca2+ transport in endoplasmic reticulum (ER) of the uterine smooth
muscle cells (SMC). Using such experimental model as suspension of
digitonin-permeabilized myometrial cells the component of
Mg2+,ATP-dependent Ca2+-accumu1ation insensitive to rethenium red,
stimulated by oxalate and phosphate, inhibited by thapsigargin and
cyclopiazonic acid was first identifiad and systematicaly investigated
by isotopic method (45Ca2+). It reflects the functioning of Ca2+-pump ER
of uterine SMC. Properties of this pump were studied: relation to ATP
and Mg2+, substrate and catione specificity; sensitivity to action of
different inhibitors of membrane-linked Ca2+-pumps and exchangers, ions
of metals. It is demonstrated that oxytocin (100 nM) partly inhibits
Mg2+,ATP-dependent Ca2+ accumulation in ER of uterine SMC.
