Lipid transport system under oxidative stress in rats. — Manuskript. The thesis is devoted to the investigation of the lipid transport system under oxidative stress development, caused by cobalt chloride or tetamisol single intraperitoneal injection. The entering of the various xenobiotics such as heavy metals ions to the human and animal organism leads to different functional and metabolic disorders, including the disorders of lipid transport and metabolism. This process is accompanied by damage of the main classes of biological macromolecules and its complexes including biological membranes also by decrease of the reduced glutathione and macroergs contents. In reply to oxidative stress a number of protective reactions directed on braking free-radical oxidation also to maintenance of functional activity of cell are formed. The various stress effectors capable to loweing a level of compounds generating the active form of oxygen and to activation antyoxidant systems induction is shown. However, the role of lipids in protective reactions under oxidative stress is not investigated enough now. Oxidative modifications of low density lipoproteins is a known aspect of atherogenesis. So the lipoprotein spectrum and composition, their oxidative status in blood serum and liver cytosol in Wistar male rats (weight 180-220 g) in various terms after single injection of cobalt chloride or tetramisol have been investigated in present work. The dissertation consists on the introduction, the review of the literature on the closen theme, the description of the main methods of the investigation, two parts of the experimental results with their discussion, conclusions and literature cited. The litrature review is devoted to the regulation of the lipid transport under extremal actions. In blood serum 5 lipoprotein fractions were investigated ((1-LP, (2-LP, (-LP, pre-(-LP and chilomicrones), and in liver cytosole — 4 fractions ((-LP, (-LP, pre-(-LP and fraction of start zone). In the experiments it has been shown that one-time intraperitonial administration of cobalt chloride or tetramisol in doses near to LD50 leads to tocopherol content in lipoprotein decreasing and lipoprotein oxidative modifications. There were no lipoperoxides in (- and pre-(-lipoproteins after long-time (24 hr) exposure howewer (-LP remain oxidatively modified. Activation of free-radical oxidation is accompanied by changes in lipoprotein content and composition. The role of (-LP peroxide modifications, rising their phospholipids content and cholesterol level in antioxidant and antiatherogenic defence is discussed. Decreasing in liver cytosole by 30 min pre-(-lipoproteins content, obviously, suggests to emission available pool of ready lipoproteins to blood. Changes in the prae-(-lipoproteins structure in liver and blood probably reflects the increasing of the tissues requirement in lipids, connected with activation of lipid peroxidation. Decrease of the total lipids, phospholipids and cholesterol ethers contents in liver cytosole start zone, probably, favours to amplification of lipid inclusion into lipoproteins. Changes of structure liver (-lipoproteins confirms an intensive lipid exchange between lipoproteins of blood and periferal tissues cells. Decreasing of the phospholipid content also in pre-(-lipoproteins in blood serum, probably, is connected to their active utilization by tissues for homeostatic correction of lipid constitution of membranes. It confirms also by the phospholipid content in liver pre-(-lipoproteins continues to increase, i.e. weakening of their inclusion to synthesized anew particles does not occur. Normalisation of total lipids content in liver pre-(-lipoproteins to 24 hr on a background of essential increasing of the fraction total content could testify about transition of biosynthetic systems of liver on new higher basal level. In liver cytosole the total lipid and lipoprotein content to 96 hr is reduced in comparison with the control that probably is connected with still intensive lipoprotein emission to blood. So the data obtained also show that the oxidative stress caused by one-time cobalt chloride or tetramisol injection leads to essential activation of lipid transport. The increase of the contents mostly all of lipoprotein fractions in blood serum and liver cytosole, and atherogenic changes (increasing cholesterol level and oxidative modifications) in lipoprotein structure were found. Total lipid content in lipoproteins was found to decrease, and hyperlipidemia is arrived at increasing in level lipid-poor lipoproteins. It has been shown that lipoprotein fractions are redistributed with increasing level of atherogenic ones. Co-administration cobalt chloride and by actinomycine D or cycloheximide (the inhibitors of the protein synthesis) has realy prevented hyperlipoproteinemia in many cases byt hasn’t influenced on lipoprotein oxidation. It has been concluded that apoprotein synthesis is very important process influenced on adaptive reactions under free-radical oxidation activation conditions. Pre-(- and (-lipoproteins were discussed to have mRNA pool in hepatocytes. In the in vitro conditions it has been found that cobalt ions directly cause lipoprotein disaggregation and free lipid level increasing in blood serum and liver cytosole. It has been concluded that system of lipids homeostasis consists of lipid synthesis and transport mechanismes in the organism is activated under oxidative stress conditions.
