The experimental and theoretical investigation of temperature influence on the platelet and fibrinogen structures under the action of (-irradiation with doses 10-250 Gy has been performed for the range of 4-44(C. The information concerning the structural changes of fibrinogen molecule at temperatures from 4 to 44(C has been obtained by means of the very high frequency dielectric spectroscopy and UV-spectroscopy methods. A new conformational transition has been observed in fibrinogen at 18-22(C. This transition might be connected to structural transition in D-domain of fibrinogen and accompanied by an increase in tryptophanyl accessibility to the solvent. The conformational transition revealed probably determines the character of the temperature dependence of blood platelet aggregation. The structural changes of platelet membrane receptors have been registered as changes of intensities of derivative maxima in the UV-range at the temperatures of 18-20(C and 30-32(C. The data on water structure change has been obtained by using very high frequency dielectric spectroscopy for the platelet suspension at 18-22(C and 32-36(C. The values of activation enthalpy for the process of dielectric relaxation of bulk waters in cell suspension have been calculated. The influence of the temperature on platelet aggregation induced by various agents has been studied by using the light scattering method. It has been shown that the temperature dependences of degree and rate of platelet aggregation have maxima. The extreme nature of the temperature dependences does not depend on the platelet count, agonist concentration, and rate of suspension stirring. The mechanism of the temperature dependence of platelet aggregation has been discussed. The existence of the membrane phase transition and change of receptor mobility allow one to explain the kink at the 20(C. The membrane proteins and fibrinogen conformational transitions take place at the same temperature. These lead to the decrease of aggregation. The mathematical model of platelet aggregation kinetics has been suggested. The case of the total platelet activation has been considered. The aggregation is modeled as the process of elastic collisions platelets and spherical aggregates. The possibility of the disassociation of platelets from an aggregate due to the breakdown of molecular bridges and collisions between aggregates. The time variations of the numbers of platelets included into an aggregate for various temperatures have been obtained as the result of the numerical simulations. The optimization of the aggregation and disaggregation rate constants has been performed using experimental data obtained with the help of light scattering method. The temperature dependences of kinetic constants have been obtained, the corresponding values of activation energies have been calculated. The temperature dependence of the aggregation platelet irradiated in vitro with doses 10-250 Gy has been studied by using light scattering method. The irradiation leads to the change of membrane viscosity and platelet protein conformation. The maximal values of aggregation have been registered for the doses 30-50 Gy. The change of the fibrinogen hydration has been registered at the same doses. The differences of the temperature dependences features of irradiated platelet aggregation induced by various agonists are connected to different radiosensitivity receptors of these agonists on the platelet membrane. It has been shown that the temperature dependence parameters of the aggregation rate and degree are sensitive to the irradiation. Prolonged combined internal and external low-dose irradiation of whole body also leads to decrease of aggregation platelet. The change of platelet activity in the dependence on the storage terms is determined by the change of membrane viscosity, cytoskeleton protein structure, and plasma aggregation co-factors. However, these changes for the long duration storage reflect the haemopoesis disturbing and the emission of the platelets with changed lipid membrane composition into circulation.
