The influence of mercury chloride and cobalt chloride on lipid peroxidation and activity of some antioxidant enzymes in rat liver, kidneys and lungs. - Manuscript. The dissertation is devoted to investigation of cobalt chloride and mercury chloride effect on lipid peroxidation intensity and some indices of antioxidant protection in liver, kidneys and lungs of Wistar rats of both sexes in different periods after injection of the salts. The influence of protein synthesis inhibitors (actinomycin D and cycloheximide) and their common use with the salts of heavy metals on lipid peroxidation intensity in rat liver, kidneys and lungs was also studied. The injection of mercury and cobalt salts was established to be accompanied by lipid peroxidation activation in liver, kidneys and lungs of male and female rats. The revealed sex peculiarities of metal salts effect on lipid peroxidation intensity lie in the fact that maximum lipid peroxidation activation in male rats was observed in half an hour after the injection and the subsequent dynamics of lipid peroxidation intensity change (in 2 and 14 or 24 hours) depended on the nature of metal. In female rats lipid peroxidation activation was maximum at the late periods after mercury chloride and cobalt chloride injection and judging by the absolute value was lower than in male rats. Considerable reduction of catalase activity was established to be already observed in liver and lungs of experimental rats in 2 hours after mercury chloride injection. There weren't essential distinctions of this activity by that time in kidneys of experimental rats. Catalase activity remained depressed in liver and lungs in 24 hours after mercury chloride injection but increased in 1,4 times in comparison with the control in kidneys. Superoxide dismutase activity was higher in lungs in 2 hours after metal salt injection, but in kidneys - in 24 hours. Superoxide dismutase and catalase activities increase in rat kidneys in 24 hours after HgCl2 injection and superoxide dismutase activity increase in lungs at early periods are explained as adaptational increase in reliability of enzymatic antioxidant system, for example, owing to induction or activation of antioxidant enzymes. Investigation of mercury and cobalt salts influence on reduced glutathione content in liver, kidneys and lungs has showed that GSH content in liver, kidneys and lungs was already decreased as compared with control in half an hour after mercury chloride injection. GSH content in all studied organs remained depressed in 24 hours after mercury chloride injection too. GSH content in half an hour after cobalt chloride injection was established to decrease only by 9% in kidneys, not to change in lungs but to increase by 14% in liver. Later on GSH content in rat liver and lungs has decreased and then slightly increased by 24 hours after injection. GSH content in kidneys of experimental rats was monotonously decreasing in the course of the experiment. At the same time the lowering of GSH content in all studied rat organs after cobalt chloride injection was less expressed than after mercury chloride injection. These data are evidence of important role of antioxidant system in maintenance of prooxidant-antioxidant balance under cobalt chloride and mercury chloride injection and permit to explain to a certain extent revealed features of lipid peroxidation activation by studied metal salts as well. Protein synthesis inhibitors (actinomycin D and cycloheximide) have activated ascorbate-induced lipid peroxidation and increased malondialdehyde content in rat liver, kidneys and lungs in 2 hours after injection. At the same time more expressed lipid peroxidation activation was recorded in rat liver under cycloheximide injection. Additional activation of lipid peroxidation in rat kidneys and lungs was established in 2 hours after common injection of actinomycin D and mercury chloride. Common injection of actinomycin D with mercury chloride or cobalt chloride didn't result in additional activation of lipid peroxidation in rat liver. Additional activation of lipid peroxidation in rat liver, kidneys and lungs wasn't found under common injection of studied metal salts with cycloheximide. The obtained data allow to suppose that lipid peroxidation activation by heavy metal salts procceds from lowering of as non-protein antioxidants content as level of different antioxidants of protein nature.
