The influence of hypoxia and nitric oxide on medium sized mice neuron calcium currents. - Manuscript. The thesis is devoted to investigation of role of the nitric oxide and it derivate - peroxynitrite in neuron hypoxic damage. The data obtained indicate that low-voltage activated (LVA) high- (HVA) and calcium currents (ICa) in 80 % of dorsal root ganglion neurons are resistant to hypoxia (pO2 = 10-20 Hg); hypoxia reduced both ICa LVA and HVA only in 20% neurons. In contradiction to hypoxia, rotenon (10 M) caused irreversible decrease of ICa in all investigated neurons. Authentic NO (10 M) reduced both LVA and HVA calcium currents. This inhibitory action was shown to be mediated substantially by an increase in cGMP level. At the same time, peroxynitrite irreversibly blocked both types of calcium currents.
